Amylin and Social Stress Amylin blunts hyperphagia and reduces weight and fat gain during recovery in socially stressed rats

چکیده
During recovery from social stress in a visible burrow system (VBS), during which a dominance hierarchy is formed among the males, rats display hyperphagia and gain weight preferentially as visceral adipose tissue. By proportionally increasing visceral adiposity, social stress may contribute to the establishment of metabolic disorder. Amylin was administered to rats fed ad libitum during recovery from VBS stress in an attempt to prevent hyperphagia and the resultant gain in body weight and fat mass. Amylin treatment reduced food intake, weight gain, and accumulation of fat mass in male burrow rats, but not in male controls that spent time housed with a single female rather than in the VBS. Amylin did not alter neuropeptide Y (NPY), agouti-related peptide (AgRP), or pro-opiomelanocortin (POMC) mRNA expression in the arcuate nucleus of the hypothalamus as measured at the end of the recovery period; nor did it effect plasma corticosterone or leptin. Amylin exerted most of its effect on food intake during the first few days of recovery, possibly through antagonism of NPY and/or increasing leptin sensitivity. The potential for chronic social stress to contribute to metabolic disorder is diminished by amylin treatment, though the neuroendocrine mechanisms behind this effect remain elusive.

متن کاملEffects of prior or concurrent food restriction on amylin-induced changes in body weight and body composition in high-fat-fed female rats.
Amylin infusion reduces food intake and slows body weight gain in rodents. In obese male rats, amylin (but not pair feeding) caused a preferential reduction of fat mass with protein preservation despite equal body weight loss in amylin-treated (fed ad libitum) and pair-fed rats. In the present study, the effect of prior or concurrent food restriction on the ability of amylin to cause weight los...
متن کاملPharmacological actions of the peptide hormone amylin in the long-term regulation of food intake, food preference, and body weight.
The ability of amylin to reduce acute food intake in rodents is well established. Longer-term administration in rats (up to 24 days) shows a concomitant reduction in body weight, suggesting energy intake plays a significant role in mediating amylin-induced weight loss. The current set of experiments further explores the long-term effects of amylin (4-11 wk) on food preference, energy expenditur...
متن کاملVMH amylin signaling is required for full leptin signaling and protection from 1 diet - induced obesity 2
23 Amylin enhances arcuate (ARC) and ventromedial (VMN) hypothalamic nuclei leptin signaling 24 and synergistically reduces food intake and body weight in selectively bred diet-induced obese 25 (DIO) rats. Since DIO I-amylin dorsomedial nucleus (DMN)-dorsomedial VMN (VMNdm) 26 binding was reduced, we postulated that this contributed to DIO VMH leptin resistance and that 27 impairing ventromedia...
متن کاملIntrathecal Amylin and Salmon Calcitonin Affect Formalin Induced c-Fos Expression in the Spinal Cord of Rats
Background: Amylin and Salmon Calcitonin belong to the calcitonin family of peptides and have high affinity binding sites in the rat spinal cord. The aim of this study was to characterize receptors for Amylin and Salmon Calcitonin functionally in the spinal cord of rats. We assessed the expression of c-Fos in response to intraplantar formalin in the lumbar regions of the spinal cord in consciou...
